Product Review: Ecotest Antigen Test

The Ecotest COVID-19 antigen rapid test is a Health Canada authorized device that uses lateral flow in vitro immunoassay for qualitative detection of the SARS-CoV-2 (COVID-19) viral nucleoprotein. This testing device detects both viable and non-viable virus from nasal swab samples.

Each test kit contains the following components:

• Test cassettes
• Nasal Swabs
• Mixing tubes pre-filled with buffer
• Mixing Tube Holder Trays

How it works:

The COVID-19 Antigen Rapid Test Device detects SARS-CoV-2 viral antigens in nasal swab samples. The test’s extraction buffer is designed to extract the SARS-CoV-2 antigens from the specimen and when a nasal swab sample is added to the buffer. The test contains a conjugate pad containing SARS-CoV-2 antibodies that specifically bind to the antigen protein. These antibodies are conjugated to colour particles which allow the binding to become visible if the antigen is detected, providing a test result that can be easily visualized.

How to take the test:

The test device should be stored at 2-30°C when not in use. Allow the test device, specimen, buffer, and/or controls to reach room temperature (15- 30°C) prior to testing.

Testing procedure

Devices, reagents and specimens and/or controls should be room temperature (15~30°C) for at least one hour prior to use.

1. For each specimen, open the foil pouch just before testing and remove the test device, and place it on a clean, level surface. Label the tube with the patient identification. For best results, the assay should be performed within one hour.

2. Remove the blue cap from the buffer fluid and place it on a safe flat surface (or in holder).

3. Obtain sample:

Bilateral Anterior Nasal Swab – insert the swab into the nostril, ~2.5cm (1-1.5″) – this is NOT far back to soft pallet, it is the main nostril cavity. Rotate the swab against the nasal wall five times you can push on the outside of your nostril to rub swab against nasal cavity to to ensure the swab has both cells and mucus.

4. Insert the swab into the extraction tube. Mix well and squeeze the swab 10-15 times by compressing the walls of the tube against the swab.

5. Let the sample sit in buffer for 2 minutes.

6. Roll the swab head against the inner wall of the tube as you remove it. Try to release as much liquid as possible. Dispose of the used swab in accordance with your biohazard waste disposal protocol.

7. Place the blue cap back onto the buffer fluid.

8. Remove the clear, upper cap, off the buffer fluid. Invert the tube and add 2 drops of solution into the sample well by gently squeezing the tube.

9. Wait for the coloured line(s) to appear. Read results in 15 minutes.

How to read the results:

A positive test result is indicated by the presence of two coloured bands on the membrane. One band appears in the control region (C) and another band appears in the test region (T).

A negative test result is indicated by the appearance of one coloured band in the control (C) region, and the absence of a coloured band in the test (T) region.
